The whole thing is about how GW501516 affects the metabolism of fatty acids. How it activates receptors responsible for regulating the expression of various genes involved in the metabolism of carbs, cholesterol and lipids as well as formation of new fat cells.

In the thesis they look at GW's effect on acetyl-CoA. If you eat more than you burn then your body produces too much acetyl-CoA and this is stored as fat. GW drastically reduces the enzyme ACC that's responsible for resulting in acetyl-CoA being stored as fat.

The data also shows that GW501516 upregulates CPT transcription. WTF is that? :blink: This facilitates the transport of fatty acids into mitachondria. These are the cells that turn fatty acids into usable energy where otherwise they could have been stored as fat.

Next it shows that GW501516 drastically reduces LPL gene activation. WTF? :blink: Lipoprotein Lipase is also involved in dreaded fat storage. Something about regulating much fatty acids are stored in adipose tissue. So reducing LPL means reducing fat storage.

The thesis also says how PPAR agonists like GW501516 (and they mention another I never heard of -GW610742) help regulate the 'reverse cholesterol transport process'. This is where our good HDL cholesterol moves bad LDL cholesterol from tissues in the body back to the liver where excess cholesterol is eliminated. Very good.

GW sounds too good to be true. ;)

think ill do 20mg tomorrow just to make sure its the gw.
not taking any other supps right now. maybe its the keto.

Finished 8 weeks @ 20mg ed. No sides to report. Only upset stomach ( constipated ) after i stopped

I mainly used it to see what the prescribed cholesterol meds do vs this GW. (awaiting blood-tests)

This may sound funny, but hey , WTF - Endurance in the bedroom was insane, i could do the midnight marathon LOLZ !

Good overall slow weight loss, endurance def a boost.

Only shitty part is when i stopped, i felt kak for 2 days. (no energy and constipated....

All good now :)

Have u all seen the new info posted on the web about gw501516. Not clear about the cancer development and if it is only on prolonged use.

Any info will help wanted to start on monday but WADA report got me wundering

GW501516, a non-genotoxic PPARδ agonist,

was assessed for carcinogenic poten- tial by

daily administration (oral gavage) to Han

Wistar rats for a period of 104 weeks. Males

were given 0, 5, 15 or 30 mg/kg/day for the

first 6 weeks of the study. For the

remainder of the study males were given 0,

5, 20 or 40 mg/kg/day. Females were given

0, 3, 10 or 20 mg/kg/day for the entire

study. GW501516 produced test article-

related neoplastic findings in multiple

tissues at all doses. Increased mortal- ity

was seen with females given GW501516 at

all doses and uterine endometrial

adenocarcinoma contributed to death in a

high proportion of these animals.

Neoplasms considered test-article related

occurred in the liver (hepatocellular ade-

noma at ≥ 10 mg/kg/day), urinary bladder

(transitional cell carcinoma in males given

20 and 40 mg/kg/day), thyroid (follicular

cell adenoma at ≥ 3 mg/kg/day and

carcinoma in males at ≥ 20 mg/kg/day),

tongue (squamous cell papilloma in males

at 5 mg/kg/day and 40 mg/kg/day), stomach

(squamous cell papilloma in males at ≥ 5

mg/kg/day and a female at 20 mg/kg/day,

and carcinoma in a male at 40 mg/kg/day

and a female at 3 mg/kg/day), skin

(inverted squamous cell papilloma in males

at ≥ 5 mg/kg/day and females at 3 or 20

mg/kg/day), Harderian glands (ade- noma

in males at ≥ 5 mg/kg/day and

adenocarcinoma in a male at 40 mg/kg/

day), testes (interstitial cell adenoma at 40

mg/kg/day), ovary (Sertoli cell adenoma at

≥ 10 mg/kg/day) and uterus (polyp and

endometrial adenocarcinoma at ≥ 3 mg/kg/

day). Some of the tumor types observed in

this study have not been reported with

either PPARα or PPARγ agonists and may

reflect tumor promotion mediated through

PPARδ agonism.


My concern about the studies I found is that 1. the "extended periods" aren't quantified as a definitive term. 2. the doses used are quite high compared to the suggested dosages. For example, 15mg/kg/day from the above study would translate to 2.4mg/kg/day in a human model. That's like 192mg/day for a 80kg person - I don't think anyone would use that much.

I'm still in 2 minds about it though and will research more before I start use.
